چکیده

Tuberculosis (TB) disease occurs in every part of the world and large numbers of cases have been noted from all over the world. About one third of the population has TB infection and millions of people are dying every year. The disease is caused by Mycobacterium tuberculosis (MTB) which usually attacks lungs but can infect other organs also. In the current situation, the major problem in treating this disease is emergence of Multidrug resistance (MDRTB) and Extensive drug resistance (XDRTB) strains, which remains a critical but largely unresolved issue. MDR-TB refers to the TB bacteria which are resistant to first line drugs such as isoniazid and rifampicin, while XDR-TB refers to the TB bacteria which are resistant to fluoroquinolone (FQ) in addition to isoniazid and rifampicin and to at least one of the second line drugs like ISSN: 2319-7706 Volume 3 Number 8 (2014) pp. 528-546 http://www.ijcmas.com

Efflux pumps of Mycobacterium tuberculosis play a significant role in antituberculosis activity of potential drug candidates.

Active efflux of drugs mediated by efflux pumps that confer drug resistance is one of the mechanisms developed by bacteria to counter the adverse effects of antibiotics and chemicals. To understand these efflux mechanisms in Mycobacterium tuberculosis, we generated knockout (KO) mutants of four efflux pumps of the pathogen belonging to different classes. We measured the MICs and kill values of ...

Antimicrobial efflux pumps and Mycobacterium tuberculosis drug tolerance: evolutionary considerations.

The need for lengthy treatment to cure tuberculosis stems from phenotypic drug resistance, also known as drug tolerance, which has been previously attributed to slowed bacterial growth in vivo. We discuss recent findings that challenge this model and instead implicate macrophage-induced mycobacterial efflux pumps in antimicrobial tolerance. Although mycobacterial efflux pumps may have originall...

Synthesis and evaluation of inhibitors of bacterial drug efflux pumps of the major facilitator superfamily.

Inhibitors of drug efflux pumps have great potential as pharmacological agents that restore the drug susceptibility of multidrug resistant bacterial pathogens. Most attention has been focused on the discovery of small molecules that inhibit the resistance nodulation division (RND) family drug efflux pumps in Gram-negative bacteria. The prototypical inhibitor of RND-family efflux pumps in Gram-n...

Novel epitopes identified from efflux pumps of Mycobacterium tuberculosis could induce cytotoxic T lymphocyte response

Overcoming drug-resistance is one of the major challenges to control tuberculosis (TB). The up-regulation of efflux pumps is one common mechanism that leads to drug-resistance. Therefore, immunotherapy targeting these efflux pump antigens could be promising strategy to be combined with current chemotherapy. Considering that CD8+ cytotoxic T lymphocytes (CTLs) induced by antigenic peptides (epit...

Involvement of efflux pumps in the resistance to peptidoglycan synthesis inhibitors in Mycobacterium tuberculosis.

We evaluated the contributions of Mycobacterium tuberculosis efflux pumps towards intrinsic resistance to different classes of peptidoglycan synthesis inhibitors (PSI). Our study indicates that the efflux pump knockout strains are more susceptible to PSI than the wild type. Vancomycin and ceftriaxone exhibited up to 3 log increased kill on efflux pump mutants compared to the wild-type strain, s...
